| The Human Cell Lineage EpiTect Chip qPCR Array profiles the histone modification status or “histone code” of 84 genes key to cellular differentiation. Histone modifications define chromatin structure, regulate gene activity and specify cellular status. During embryonic development, pluripotent stem cells differentiate into three germ layers: ectoderm, mesoderm and endoderm. These germ layers eventually differentiate into multipotent stem cells (progenitors), which progress into terminally differentiated cells. These developmental processes require tightly regulated and carefully timed gene expression changes. Epigenetic histone modifications are one such mechanism of regulation, where gene promoters are activated, repressed, or "poised" in an intermediate state. This array contains gene markers for specific cell types throughout cellular lineage progression, including pluripotent stem cells, progenitor cells from each of the three germ layers, and terminally differentiated cells. Surveying the histone modification pattern of these promoters can help explain the observed up- or down-regulation of their expression, thereby elucidating differentiation mechanisms, and can help validate the correct epigenetic status of differentiation programs and terminally differentiated cells. Using chromatin immunoprecipitation and this real-time PCR Array, you can easily and reliably analyze the histone modification patterns associated with a focused gene panel of cellular lineage progression markers.
